Chemical investigation on gorgonian coral Subergorgia suberosa collected from the South China Sea led to the isolation and identification of eight 17-acetyl pregnane steroids (18), of which 1, 6 and 7 were obtained for the first time from this species, as well as from the genus Subergorgia. Compounds 1 and 7 are new natural products. Compared with other species in the genus Subergorgia, pregnane steroids with 17-acetyl group (18) were only achieved from S. suberosa, suggesting that the 17-acetyl pregnane steroids might be characteristic constituents of S. suberosa, and may be useful as chemotaxonomic markers for the species S. suberosa.  相似文献

Synchronized state of activity and rest might be attained by mechanisms of entrainment and masking. Most zeitgebers not only act to entrain but also to mask circadian rhythms. Although the light-dark (LD) cycle is the main zeitgeber of circadian rhythms in marmosets, social cues can act as weaker zeitgebers. Evidence on the effects of social entrainment in marmosets has been collected in isolated animals or in pairs where activity is not individually recorded. To characterize the synchronization between the daily activity profiles of individuals in groups under LD conditions, the motor activity of animals from five groups was continuously monitored using actiwatches for 15 days during the 5th, 8th, and 11th months of life of juveniles. Families consisting of twins (4 ♂♀/1 ♂♂) and their parents were maintained under controlled lighting (LD 12:12?h), temperature, and humidity conditions. Synchronization was evaluated through the synchrony between the circadian activity profiles obtained from the Pearson correlation index between possible pairs of activity profiles in the light and dark phases. We also calculated the phase-angle differences between the activity onset of one animal in relation to the activity onset of each animal in the group (ψon). A similar procedure was performed for activity offset (ψoff). By visual analysis, the correlation between the activity profiles of individuals within each family was stronger than that of individuals from different families. A mixed-model analysis showed that within the group, the correlation was stronger between twins than between twins and their parents in all families, except for the family in which both juveniles were males. Because a twin is an important social partner for juveniles, a sibling is likely to have a stronger influence on its twin’s activity rhythm than other family members. Considering only the light phase, the second strongest correlation was observed between the activity profiles of the individuals in the reproductive pair. Regarding the parameters ψon and ψoff, the juvenile/juvenile dyad had lower values than the other dyads, but these differences did not reach statistical significance in relation to all dyads. Comparing the results of the ψon and ψoff, and correlation indices, we suggest that the latter could detect differences between the animals that were not observed in the results of the phase-angle differences. These differences could be related to changes that occur during the active phase but not only in a particular phase, such as the temporal changes during the activity phase that characterize unimodal or bimodal patterns. Based on the differences in the correlations between individuals subjected to the same LD routine, we suggest that social cues modulate the circadian activity profiles of marmosets as a result of interactions between the animals within each group. Abstract

Metal ions binding to proteins regulate the functions of proteins and may also lead to structural changes. In this communication we demonstrate the interaction and subsequent conformational changes induced in pig gastric mucin (PGM) upon binding to certain chromium(III) complexes like, [Cr(salen)(H2O)2](ClO4) (1), [Cr(en)3]Cl3 (2) and [Cr(EDTA)(H2O)]Na (3) which vary in charge and ionic character. Complexes 1 and 3 have been shown to interact coordinately with PGM whereas complex 2 binds through electrostatic interaction and hydrogen bonding. Steady state fluorescence experiment reveals that at lower concentration of complex 2 there is partial quenching of the tyrosine emission, whereas at higher concentration of the complex the emission intensity is enhanced. On the other hand with complexes 1 and 3 a decrease in fluorescence intensity was observed. PGM viscosity was found to decrease in the presence of complex 1 and 3 due to the formation of flexible fibres through coordinate interaction. Complex 2 was found to facilitate metal induced intertangling of PGM fibres which tends to stabilize the interaction and leads to sol-gel transition with subsequent increase in viscosity. A significant change in CD spectrum of PGM was observed in the presence of complex 2 where random coil spectrum became typical of a α-helical structure with 80% alpha helix content. In the case of complexes 1 and 3 only minor changes in the amplitude of the spectrum were observed. Histochemical analysis supports the contention that complex 2 favors the oligomerisation of PGM and leads to the formation of aggregated mass of macromolecules.  相似文献

The ABO histo-blood group antigens are best known for their important roles in solid organ and bone marrow transplantation as well as transfusion medicine. Here we report the synthesis of the ABO type III and IV antigens with a 7-octen-1-yl aglycone. Also described is an NMR study of the ABO type I to VI antigens, which were carried out to probe differences in overall conformation of the molecules. These NMR investigations showed very little difference in the 1H chemical shifts, as well as 1H–1H coupling constants, across all compounds, suggesting that these ABO subtypes adopt nearly identical conformations in solution.  相似文献

Large numbers of hatchery-reared fish are released in stocking programmes; however, success is limited by high mortality. Predation is seen as the main cause of deaths but might be reduced by training fish before release to avoid predators and/or use refuge. In this study on a potential restocking species, yellowfin bream Acanthopagrus australis, the effects of predator training and refuge on the behaviour of fish in the hatchery were tested. In the first experiment, juvenile bream were exposed to predatory mangrove jack (Lutjanus argentimaculatus) fed exclusively on bream flesh while housed in tanks with and without refuge. Predator training altered fish behaviour when fish were re-exposed to predators, but the effects were subtle and varied between groups of fish. In contrast, refuge created strong and consistent changes in behaviour, significantly slowing down the amount of time that fish took to consume food. A second experiment focused on the effects of refuge. Bream were trained to use artificial seagrass or house bricks as refuge and then exposed to mangrove jacks in a laboratory predation experiment. When refuge was available, fish significantly slowed down their feeding rate. There was a small, transient increase in survival for fish given seagrass refuges, but this was irrespective of whether the bream were trained to use refuge. The results of this study indicate that the use of refuge may be innate and the benefits of refuge may be available to naive hatchery-reared fish or fish trained to use refuge shortly before release. This suggests that there is potential to improve post-release survival of fingerlings without time-consuming and expensive hatchery training.  相似文献

Many organisms exhibit phenotypic plasticity; producing alternate phenotypes depending on the environment. Individuals can be plastic (intragenerational or direct plasticity), wherein individuals of the same genotype produce different phenotypes in response to the environments they experience. Alternatively, an individual's phenotype may be under the control of its parents, usually the mother (transgenerational or indirect plasticity), so that mother's genotype determines the phenotype produced by a given genotype of her offspring. Under what conditions does plasticity evolve to have intragenerational as opposed to transgenerational genetic control? To explore this question, we present a population genetic model for the evolution of transgenerational and intragenerational plasticity. We hypothesize that the capacity for plasticity incurs a fitness cost, which is borne either by the individual developing the plastic phenotype or by its mother. We also hypothesize that individuals are imperfect predictors of future environments and their capacity for plasticity can lead them occasionally to make a low‐fitness phenotype for a particular environment. When the cost, benefit and error parameters are equal, we show that there is no evolutionary advantage to intragenerational over transgenerational plasticity, although the rate of evolution of transgenerational plasticity is half the rate for intragenerational plasticity, as predicted by theory on indirect genetic effects. We find that transgenerational plasticity evolves when mothers are better predictors of future environments than offspring or when the fitness cost of the capacity for plasticity is more readily borne by a mother than by her developing offspring. We discuss different natural systems with either direct intragenerational plasticity or indirect transgenerational plasticity and find a pattern qualitatively in accord with the predictions of our model.  相似文献

Tuberculosis killed 1.5 million people in 2018. Mycobacterium tuberculosis (Mtb), the causative agent of tuberculosis, is the most deadly infectious bacteria in the world. A strength of mycobacterial pathogens — their formidable cell wall — could also be one of their greatest molecular vulnerabilities. As in other bacteria, peptidoglycan (PG) maintenance and integrity is essential to mycobacterial survival. But Mtb PG is unique, and a better understanding of its biosynthetic machinery could lead to new drugs or more effective treatment regimens. Such investigations are being accelerated by the application of fluorescent probes, including those based on vancomycin, β-lactams, PG stem mimics, d-amino acids, and reactive glycans. This review will describe how fluorescent probes are being used to uncover new information on the regulation and drug susceptibility of two classes of enzymes that fortify the Mtb PG: the penicillin-binding proteins and the L,D-transpeptidases.  相似文献

The decoration of proteins with post-translational modifications (PTMs) serves as a mechanism to expand the functional repertoire of the proteome. Tyrosine sulfation is a PTM that has been shown to be a key regulator of extracellular protein–protein interactions in a select number of examples. However, the challenges associated with identifying and characterising the functional consequences of tyrosine sulfation have hindered our ability to understand the full scope of its role in the wider proteome when compared with that of other PTMs, for example, phosphorylation and glycosylation. In this account, we highlight recent advances in the prediction and detection of tyrosine sulfation and outline the need for continued innovation in this area. We also discuss the utility of emerging solid-phase synthesis and peptide ligation strategies for accessing libraries of homogeneously sulfated peptides and proteins to help reveal functional aspects of the sulfoproteome.  相似文献

Formants are important phonetic elements of human speech that are also used by humans and non-human mammals to assess the body size of potential mates and rivals. As a consequence, it has been suggested that formant perception, which is crucial for speech perception, may have evolved through sexual selection. Somewhat surprisingly, though, no previous studies have examined whether sexes differ in their ability to use formants for size evaluation. Here, we investigated whether men and women differ in their ability to use the formant frequency spacing of synthetic vocal stimuli to make auditory size judgements over a wide range of fundamental frequencies (the main determinant of vocal pitch). Our results reveal that men are significantly better than women at comparing the apparent size of stimuli, and that lower pitch improves the ability of both men and women to perform these acoustic size judgements. These findings constitute the first demonstration of a sex difference in formant perception, and lend support to the idea that acoustic size normalization, a crucial prerequisite for speech perception, may have been sexually selected through male competition. We also provide the first evidence that vocalizations with relatively low pitch improve the perception of size-related formant information.  相似文献
